چکیده

Abstract Background Glioblastoma multiforme is a highly malignant primary brain tumor with an average survival of 14 months and very limited therapeutic options. Glioma associated microglia macrophages (GAMs) foster growth by releasing several cytokines, which have only partly been identified. Here, we studied the chemokine (C-C motif) ligand 18 (CCL18), expressed in human, but not rodent GAMs, novel ex-vivo slice model including transplantation human induced pluripotent stem cells (iPSC) derived (iMGL) glioma to murine slices, had depleted intrinsic before. Material Methods After establishing humanized model, performed immunohistochemical analysis (IHC) invasiveness, qrtPCR on isolated magnetic-activated cell sorting (MACS), functional assays measuring proliferation, migration colony formation vitro experiments. Corresponding studies invasiveness were after treatment CCL18 neutralizing antibody, CCR8 antibodies knockdown CCR8, ACP5 (Acid Phosphatase 5) PITPNM3 small interfering RNA (siRNA) short hairpin (shRNA). QrtPCR, IHC Westernblot specimens. We also conducted bioinformatic analyses, based TCGA GBM, GLIOVIS GEPIA databases. Results observed that was whereas cells. identified receptor 8 (CCR8) as for important down-stream signaling component Activation CCL18/CCR8/ACP5 pathway glioblastoma enhanced invasiveness. Conclusion GAMs promoted activation CCR8/ACP5 axis therefore potential target.

Identification of human CCR8 as a CCL18 receptor

The CC chemokine ligand 18 (CCL18) is one of the most highly expressed chemokines in human chronic inflammatory diseases. An appreciation of the role of CCL18 in these diseases has been hampered by the lack of an identified chemokine receptor. We report that the human chemokine receptor CCR8 is a CCL18 receptor. CCL18 induced chemotaxis and calcium flux of human CCR8-transfected cells. CCL18 bo...

S100B promotes glioma growth through chemoattraction of myeloid-derived macrophages.

PURPOSE S100B is member of a multigenic family of Ca(2+)-binding proteins, which is overexpressed by gliomas. Recently, we showed that low concentrations of S100B attenuated microglia activation through the induction of Stat3. We hypothesized that overexpression of S100B in gliomas could promote tumor growth by modulating the activity of tumor-associated macrophages (TAM). EXPERIMENTAL DESIGN...

P-52: Brain-Derived Neurotrophic Factor Promotes The Development of Human Ovarian Early Follicles during Growth In Vitro

Background Cryopreservation of ovarian cortex is increasingly used to preserve fertility before cancer therapy. Recently, studies show that Brain-derived neurotrophic factor (BDNF) may be involved in oocyte maturation. Brain-derived neurotrophic factor (BDNF) is member of neurotrophin family that has anti-apoptotic effects on nervous system. Recent researches show that it also plays key role in...

Platelet-derived growth factor in human glioma.

Platelet-derived growth factor (PDGF) is a 30 kDa protein consisting of disulfide-bonded dimers of A- and B-chains. PDGF receptors are of two types, alpha- and beta-receptors, which are members of the protein-tyrosine kinase family of receptors. The receptors are activated by ligand-induced dimerization, whereby the receptors become phosphorylated on tyrosine residues. These form attachment sit...

β-catenin-mediated YAP signaling promotes human glioma growth

BACKGROUND Hippo/YAP pathway is known to be important for development, growth and organogenesis, and dysregulation of this pathway leads to tumor progression. We and others find that YAP is up-regulated in human gliomas and associated with worse prognosis of patients. However, the role and mechanism of YAP in glioma progression is largely unknown. METHODS The expression of YAP in glioma tissue...
